Isolation,Identification and Performances of an Oxytetracycline-Degrading Strain

Abstract
A strain named OTC-1 was acclimated and isolated from solid wastes from a large scale farm in Hunan Province, and it was found that it could efficiently degrade oxytetracycline.Morphological observations and a comparative analysis of the 16SrDNA gene sequence discovered that the bacterium was 99% similar to Klebsiella pneumonia sp..This bacterium can use oxytetracycline as the only carbon source and grow in shaking cultivation. The UV spectrum was used to analyze the growing characteristics of the bacterium, while the high performance liquid chromatography (HPLC) was used to determine the changes of oxytetracycline concentration. The results showed that the degradation rate of oxytetracycline by OTC-1 reached 76.73% in mineral culture, under the condition of 150mg/L of oxytetracycline, 37℃, pH 7.0, bacterium inoculum load of 10% and shaking speed at 140r/min. The research indicated that the degradation rate of oxytetracycline was significantly increased by OTC-1.
